To mark its 100th episode, The Big Bang Theory is trying a first — and it has Kaley Cuoco slightly nervous.

“The episode is a little different from what we’ve done. It plays a misguided trick on the audience until the end, so people might be confused, but it’s very well done,” she tells TVGuide.com. “I hope people like it! The audience that we shot in front of loved it, so that’s a good start.”

Titled “The Recombination Hypothesis,” the milestone episode centers on the relationship that started the hit series: Leonard (Johnny Galecki) and Penny (Cuoco). While calling back to the pilot — when Leonard first laid eyes on Penny, of course — the installment will explore a possible reconciliation between the two after he asks her out to dinner. The promo, as seen below, shows the former couple, who broke up in Season 3 when Penny didn’t return his “I love you,” locking lips — but that may or may not be real.

“It’s one of those what-ifs,” Cuoco says. “We see what they’re both thinking a little bit and what everyone’s feeling. They’re both kinda like, ‘If we do go on this date, what will happen?’ They both build it up in their minds and then you find out in the end what’s really going on, but there are definitely a lot of questions throughout. It was extremely smart and well-written and witty. It’s a slightly different angle, and after five and a half seasons, you have to take some chances.”

Because of the various scenarios floating around in their heads, nothing really happens in the episode until the end reveal, Cuoco says. That will set up the following episode, which will offer more insight into whether Penny and Leonard get back together. “They’re not done,” Cuoco teases. “The dinner is going to be a good jumping-off point to what they’re going to start and try doing.”

With Penny having drunkenly admitted to Raj (Kunal Nayyar) that she regretted breaking up with Leonard, would it be fair to assume that she wants to start dating again? “I think even if she wants to, there are a lot of things to [consider] and it’s definitely not easy for them to just start dating again,” Cuoco says. “Their friendship is finally going well. I think what people can expect is that they’re a little older now, slightly more mature, so there won’t be some crazy, horrible drama. They each understand where the other stands.”

Still, Cuoco — who does not know the writers’ plans for Leonard and Penny — believes that it will “take awhile” for the duo to work everything out and truly reunite. And when they do, she hopes it’s for good.

“I do hope they end up together. I don’t see the show ending any time soon, so it would probably be awhile down the road,” she says. “I enjoy the back and forth. It’s extremely realistic, especially when two people live next door and have dated and have loved each other. You break up and you have to deal with your friends and think, ‘I don’t know. I may still love this person.’ Leonard is very special to Penny and vice versa. I think they foresee that this could be a life-changing relationship, but I think that’s also why they’re moving slowly with it and are looking back and ahead.”

As far as looking back goes for Cuoco, that’s one thing she doesn’t want to do — when it comes to the pilot at least. Executive producers Chuck Lorre, Bill Prady and Steve Molaro re-watched the pilot before writing the 100th episode, but Cuoco made sure to steer clear of it.

“Oh, no. I’ve seen that thing way too many times!” she says. “It’s a great pilot — I’m so proud of it and I love that [this episode] takes us back to it— but I can remember every outfit I wore in that. I don’t need to see that again for a while! Maybe by the 200th!”

We won’t sing the whole song for you (although we’re proud to say we can) we just wanted to get you pumped up in anticipation for tonight’s 100th episode of The Big Bang Theory! We interviewed our favorite geek-chic cast at their 100th episode bash and we’ve got all the scoop on tonight’s “quirky” and deceitfully fun plot twists…

Creator Chuck Lorre revealed that the audience can expect to see an homage to the pilot in tonight’s episode. “Well, we’re kind of circling back to where we started with Leonard and Penny. That romance is where we started and it felt right to kind of come back to that in the 100th episode.”

There are plenty of small touches throughout the episode that are going to make even the most devoted BBT fans feel appreciated. Fun fact: When Leonard comes in to ask Penny on another “first date” she is wearing the same shirt as the pilot when he first laid eyes on her across the hall. Awww!

“The writers wanted to do something special for the 100th and I think they accomplished that,” the always-adorable Kaley Cuoco told us. “It’s like one we’ve never done before. It’s kind of a mind game. You won’t figure it out until the end.”

Golden Globe nominee Johnny Galecki echoed her thoughts saying there are a lot of “layers” to this episode. “There’s a big twist, a bigger twist than I think we’ve ever done on this show.” Galecki continues. “It’s a departure for us, there’s a lot of nearby parallel universes and fantasies.”

Melissa Rauch gave us more of an explanation: “It’s kind of a journey with Leonard and Penny and revealing what may happen with them and the rest of the cast is looking on.”

Keyword here being “may.” Let’s put our speculation caps on, shall we? We’re guessing that this episode is going to feature some alternate parallel universe in which Leonard and Penny explore their feeling for each other but in the end it’s not the reality that we’ve been watching for the past 100 episodes. Deep, huh?

But if Leonard and Penny are going to be out on another “first date,” what is Sheldon going to be up to? Fixating on a life-size cardboard cutout of Spock of course!

“I think Sheldon is up to being typical Sheldon; he’s not making any great strides in that episode,” Jim Parsons tells us with a smile. “I spend a decent amount of time playing a certain board game with Wolowitz and Koothrappali and it was a nice just “staying the course” episode for me.” And after the excitement and perfection of last week’s first Shamy kiss, fans will probably be all right with Sheldon’s normal neurotics.

Whatever happens in tonight’s episode is sure to be a memorable experience and something that everyone will be talking about tomorrow. Galecki sums it up: “I can tell you it’s one of my favorite episodes. It might be our proudest moment.”

Thursday’s 100th episode of The Big Bang Theory is almost here and we have absolutely nothing to tell you…aBazinga! Of course we do! Kaley Cuoco revealed to us, “It’s like nothing we’ve ever done before, it’s kind of a mind game and you wont figure it out until the end, it’s really interesting, it’s special, it’s different.” The Leonard and Penny relationship will be “teased” in this episode and Cuoco says she’s keeping her fingers crossed they’ll give it another go, “At the end of the day I want it to work and I think everyone wants to know the love of their life lives next door.” Awww!

The nominees for the 2012 Screen Actors Guild Awards were announced this morning and “The Big Bang Theory” has been nominated for “Outstanding Performance by an Ensemble in a Comedy Series”!

The 18th annual SAG Awards show will be televised live from Los Angeles January 29th.

The Producers Guild of America this morning unveiled the TV series nominees for the 23rd annual PGA Awards, in which the winners will be announced January 3rd. The awards will be handed out January 21st at a ceremony that will also dole out special honors to producers extraordinaire Steven Spielberg, Leslie Moonves and Stan Lee, among others.
